Zerit Information

Common use

Zerit contains stavudine which is a synthetic nucleoside analogue of thymidine. After penetration to a cell stavudine turns into its active metabolite stavudine triphosphate, which inhibits HIV reverse transcriptase competing with the natural substrate deoxythymidine triphosphate and impairs the replication of HIV. Stavudine triphosphate also increases viral DNA chain termination due to the absence in the molecule Z'-hydroxyl groups important for the construction of DNA. In addition, stavudine triphosphate suppresses cellular DNA polymerases β and γ and considerably reduces the synthesis of mitochondrial DNA. In combination with other nucleoside and non-nucleoside reverse transcriptase inhibitors and inhibitors of HIV protease Zerit is used to treat HIV infections.

Dosage and direction

Dosage depends on body weight: for patients with body weight more than 60 kg 40 mg of Zerit is recommended every twelve hours; for patients with body weight under 60 kg 30 mg of Zerit should be taken every twelve hours. If you have swallowing problems open the capsule and take its content with some food. Dose reduction is recommended in patients with kidney impairments. Take with a full glass of water with or without food.

Precautions

Cautiousness should be exercised in patients with alcoholism, chronic renal insufficiency, liver failure, peripheral neuropathy, pancreatitis, in patients concomitantly treated by didanosine. Inform your doctor if you are pregnant or breastfeeding. Breastfeeding during treatment with Zerit should be avoided. This medication may cause fatal conditions associated with damage to the liver. If you experience fast heart rate, nausea and vomiting, severe pain in the upper stomach spreading to your back, low fever, diarrhea, loss of appetite, jaundice, dark urine, clay-colored stools. Inform your doctor if you have liver or kidney disease, a history of pancreatitis or have allergy to other antiviral drugs such as lamivudine (Epivir), tenofovir (Viread), abacavir (Ziagen), zalcitabine (Hivid), didanosine (Videx), or zidovudine (Retrovir). Some patients may develop acidosis while on Zerit. Its symptoms which you should immediately inform your doctor about even if they are mild or moderate are numb or cold feeling in the estremities, muscle pain or weakness, trouble breathing, fast or uneven heart rate, dizziness, stomach pain, nausea with vomiting, or feeling very weak or tired.

Contraindications

Zerit cannot be used in the patients with hypersensitivity to stavudine and other components of the drug, and also in children under 3 months of age. Side effects The most frequent side effects are a decrease in blood cells, muscle pain (myopathy), pancreatitis, liver failure and metabolic disturbance (lactic acidosis), a severe peripheral neuropathy (numbness and pain in the feet or hands, tingling, abdominal pain, weight loss, chills, rash, and insomnia)

Drug interaction

Inform your doctor if you take such drugs as doxorubicin (Adriamycin), didanosine (Videx), hydroxyurea (Droxia, Hydrea), ribavirin (Copegus Virazole, Rebetol, Ribasphere), interferon-alfa (Roferon, Intron, Rebetron), zidovudine (Retrovir).

Missed dose

If you forgot to take your dose in time, please do it as soon as you remember. But do not take if it is too late or time for your next dose is in a short while. Do not take double doses of this medication or extra doses of it. Take your usually dose next day in the same regularly time.

Overdose

Seek immediate medical attention if you suppose overdose of Zerit and experience tingly feeling, nausea, numbness, loss of appetite, burning, pain, stomach pain, low fever, dark urine, clay-colored stools, or jaundice.

Storage

Store at room temperature between 20-25 C (68-77 F). Store away from moisture, heat, and sunlight. It is not recommended to store in a bathroom and places available for children.
